Description: This transcription-competent virus-like particle is comprised of the Bundibugyo ebolavirus Matrix protein VP40, the secondary Matrix protein VP24, the ebolavirus Glycoprotein GP, and the ebolavirus Nucleoprotein NP. The VLP encapsidates an ebolavirus minigenome expressing the reporter protein nano Luciferase (nLuc), as well as the other proteins of the Ribonucleprotein complex (Polymerase protein L, Transcription Activator protein VP30, and Polymerase cofactor protein VP35).
Strain History: Bundibugyo ebolavirus emerged as a distinct species of ebolavirus during a 2007 outbreak in western Uganda. The genomic sequence in available in NCBI.

Production: Co-transfection of plasmids expressing the nLuc minigenome, structural proteins (VP40, GP, NP, VP24) and ribonucleoprotein complex (L, VP30, VP35) into human embryonic kidney 293T (HEK 293T) cells. Collected supernatant purified and concentrated by sucrose gradient centrifugation.

Formulation: 1.0 mg of protein suspended in PBS.
Format: 1.0 mL/vial. Purified and concentrated cell culture supernatant frozen at -80°C. Delivered on dry ice. Additional kit components include L, VP30, VP35 and NP protein expressing plasmids.
Instructions For Use: Prior to infection of cells with VLPs, transfect cells for 24-48 hours with the L, VP30, VP35 and NP protein expressing plasmids provided in the kit using a suitable transfection reagent.

Ebolavirus Minigenome Construction: The negative-sense ebolavirus minigenome encoding nLuc consists of the authentic viral leader and trailer UTR sequences at the 3' and 5' ends, which retain the essential cis-acting promoter signals required for genome transcription. Sandwiched between these terminal UTRs is a negative-sense nLuc open reading frame flanked by NP gene 5' UTR start and L gene 3' UTR end sequences.
